The theory stating that oxidative stress is at the root of several diseases is extremely popular. However, so far, no antioxidant is recommended or offered by healthcare systems neither approved as therapy by regulatory agencies that base their decisions on evidence-based medicine. This is simply because, so far, despite many preclinical and clinical studies indicating a beneficial effect of antioxidants in many disease conditions, randomised clinical trials have failed to provide the evidence of efficacy required for drug approval. In this (...) review, we discuss the levels of evidence required to claim causality in preclinical research on OS, the weakness of the oversimplification associated with OS theory of disease and the importance of the narrative in its popularity. Finally, from a more translational perspective, we discuss the reasons why antioxidants acting by scavenging reactive oxygen species might not only prevent their detrimental effects but also interfere with essential signalling roles. We propose that ROS have a complex metabolism and are generated by different enzymes at diverse sites and with different timing. Aggregating this plurality of systems in a single theory of disease may not be the best way to develop new drugs, and future research may need to focus on specific oxygen-toxifying pathways rather than on non-specific ROS scavengers. Finally, similarly to what is nowadays required for clinical trials, we recommend making unpublished data available in repositories, as this will allow big data approaches or meta-analyses without the blinders of the publication bias. What is the Critique of Pure Reason about? The terminology of the work is so perplexing, its argument so obscurely expressed, that the ordinary reader may be forgiven if he puts it down at the end very much in the dark as to what it all means. He will have seen that in it Kant has attempted to establish certain conclusions: the subjectivity of space and time, the existence and objective validity of a number of a priori concepts or categories, (...) the falsity of the arguments used to defend the metaphysical system most widely favoured in German learned circles in the eighteenth century; but though he has grasped all this he may yet have failed to make sense of the work as a whole. It is the old story of not seeing the wood for trees; and in this case the fault is more excusable than in most, for the individual trees each demand so much attention and are so difficult to get round that it is all too easy to forget the very existence of the wood. At the worst, one may think that there is no wood at all; only a miscellaneous aggregate of individual trees which have nothing to do with each other. (shrink)

Originally published in 1965 and written by one of the world's leading experts in animal behaviour, this book was written just as the impact of DNA on biology, genetics ethology and biophysics was being felt. The book reviews these developments and analyses the affect they have on our view of our own nature and of ethical and moral sense. It is particularly concerned with the impact of DNA and genetics on philosophic thought.

Few major philosophers show evidence of having studied the works of their predecessors with special care, even in cases where they were subject to particular influences which they were ready to acknowledge. Hume knew that he was working in the tradition of ‘some late philosophers in England, who have begun to put the science of man on a new footing’—‘Mr Locke, my Lord Shaftsbury, Dr Mandeville, Mr Hutchinson, Dr Butler, &c.’ But there is not much sign in the Treatise or (...) elsewhere in Hume's writings of any close acquaintance with the works of these authors; the presumption must be that he had read them at some time and extracted the main ideas, but was not in the habit of returning to their texts. He had something more important to do, namely to work at philosophical problems of his own. Similarly Kant, though he said that the Critique of Pure Reason was not meant to be ‘a critique of books and systems, but of the faculty of reason in general’, had clearly felt the impact of the thought of some important past philosophers, but equally had never spent much time in finding out just what these philosophers had to say. Plato, Aristotle, Descartes, Locke, Leibniz and Hume all get fairly frequent mention in his pages. But Kant takes his knowledge of Plato and Aristotle from J. J. Brucker's Historia critica philosophiae, a six-volume compilation which first appeared in 1742, or from doubtful sources such as Mendelssohn's doctored translation of the Phaedo, and though he doubtless knew the more recent authors at first hand clearly felt no need to study them in any depth. This was true even of writers to whom he attributed a particular importance, such as Leibniz and Hume. The references to Hume in the Critique and Prolegomena are all disappointingly general, and though the summary of Leibniz's philosophy in the section called ‘The Amphiboly of Concepts of Reflection’ has a certain force, it is not documented with references to Leibnizian texts. Kant knows that there is a difference between the views of the historical Leibniz and those which constituted the ‘Leibnizian-Wolffian system’ of his successors. But he is not very curious about the difference, or inclined to explore it. (shrink)

So much for the Aesthetic. We can now proceed to the Analytic, the philosophical importance of which is much greater. Kant's main contentions in this part of his work can be summed up in; two propositions: human understanding contains certain a priori concepts, and on these are based certain non-empirical principles; these concepts are only general concepts of a phenomenal object, and therefore the principles in question are only prescriptive to sense-experience. As has already been said, interest in the first (...) proposition has distracted attention from the fact that the important thing Kant has to say is contained in the second. (shrink)
